Use
① The intermediate of fosfomycin, can also be used as vulcanization accelerator, cosmetics (cream, lotion), mineral oil, paraffin emulsifier, biological buffer.
② Used as acid gas absorbent, preparation of buffer, surfactant, emulsifier and accelerator. It is also used in organic synthes.
Production Method
Dissolve Tris with a small amount of double steaming water (300-500ml), add HCl, adjust pH to 7.6 with HCl (1N) or NaOH (1N), and finally add the double steaming water to 1000ml. This liquid is the reserve liquid, stored in the refrigerator at 4℃.
Note: The PH value of Tris-Hcl varies with temperature, so it should be measured at room temperature, so the results measured are more reliable.
Buffering characteristic
Tris is a weak base with a pKa of 8.1 at room temperature (25℃). According to buffering theory, the effective buffering range of Tris buffers is between pH7.0 and 9.2.
The pH of the aqueous solution of Tris base is about 10.5, and the buffer of the pH value can be obtained by adding hydrochloric acid to adjust the pH value to the desired value. However, attention should be paid to the influence of temperature on the pKa of Tris. Degree, PH value decreased by 0.03.
1M Tris-HCl 6.8 and 1.5M Tris-HCl 8.8 are the most commonly used reagents for SDS-PAGE.
While TAE, TBE and other reagents synthesized from Tris are the most commonly used reagents for DNA electrophoresis, TE (pH8.0) is mainly used for DNA dissolution. (TE is Tris plus EDTA.)
